- 在4 ℃且无外加基质条件下,将实验室长期培养的硫自养反硝化颗粒污泥储存60 d,然后取出进行50 d的再生实验,探究颗粒污泥反硝化活性及形态的快速恢复机制。经过60 d的低温储存,污泥颗粒大部分解体,对硝态氮的去除率仅为原来的20.26%。经过3个阶段的恢复培养,颗粒污泥的脱氮效果基本恢复至储存前,氮去除负荷最高达到1.59 kg/(m3·d);颗粒污泥的平均粒径为0.658 mm;反应器内污泥生物量提高,污泥沉降性能良好(SVI30/SVI5=1),胞外聚合物含量明显提高,其中蛋白质(PN)在颗粒污泥的形成和维持过程中发挥着重要作用;高通量测序结果表明,硫自养反硝化菌属(Sulfurimonas和Thiobacillus)的相对丰度达到79.20%,与储存前的水平接近。低温储存60 d后的硫自养反硝化颗粒污泥,接入UASB反应器中可在50 d内基本恢复原有活性及形态。
- Abstract:
- The sulfur-based autotrophic denitrifying granular sludge, which was cultured in the laboratory over an extended period, was stored for 60 days at 4 ℃ in the absence of external substrate. Subsequently, it was subjected to a 50-day regeneration experiment to investigate the rapid recovery mechanisms of both denitrifying activity and granular morphology. After 60 days of low-temperature preservation, the majority of the granules had disintegrated, resulting in a nitrate nitrogen removal efficiency that was only 20.26% of the initial value. After undergoing three stages of recovery, the nitrogen removal performance of granular sludge was largely restored to its pre-storage level, with the maximum nitrogen removal load reaching 1.59 kg/(m3·d). The mean particle size of the granular sludge was determined to be 0.658 mm. The biomass of sludge in the reactor increased, exhibiting excellent sludge settling performance (SVI30/SVI5=1). Additionally, the content of extracellular polymeric substances (EPS) significantly rose, with protein (PN) playing a crucial role in the formation and stabilization of granular sludge. The high-throughput sequencing results demonstrated that the relative abundance of sulfur-based autotrophic denitrifying bacteria, specifically Sulfurimonas and Thiobacillus, reached 79.20%, a value nearly equivalent to the pre-storage level. The sulfur-based autotrophic denitrifying granular sludge stored at low temperature for 60 days was able to largely restore its original activity and structure within 50 days following the restart of the UASB.
