- Gold immunochromatography colloidal assay (GICA) is the only technique available to detect microcystin (MC)-LR on-site. However, there are some problems in field application of GICA, such as the inability to detect MC-LR in algal cells, the change of detection limit with ambient temperature and matrix effect. To facilitate the on-site investigation and early warning of the total MC-LR (TMC-LR) at water sources, the pretreatment method of boiling, concentration, PES membrane filtration and pH adjustment (optional) was developed and the related parameters were optimized. The limits of pH, protein and polysaccharide tolerated by the test card were 2-12, 2 g/L and 200 g/L, respectively. The optimum boiling time was 30 min, and the detection limit of 1 μg/L at ambient temperature of 10-37 ℃ could be ensured by 1 time concentration. Oocystis was used to simulate the early warning stage of algal bloom in a water body, the technical performance of the method was verified by using simulated and real water samples, and the results met the requirements of selection and detection, indicating that the method could be used for rapid detection of TMC-LR in surface water.
